Advantage

  • Operation time is very short ( <20 minutes).

Disadvantage

  • We have to centrifuge the lysate mixture twice i.e. cumbersome.
  • We need additional collection tubes ( one time but provided in an extraction kit).
  • Sufficient lysis of protein needs 1o minutes incubation at room temperature of the sample mixture.
  • To make operation time very short, dry membrane step, centrifuge at 18,000 ×g for 1 minute was adjusted but this typing speed covering centrifuge is hard to get in the common laboratory.
  • If you are using a master kit of the same company (GB), you should add internal control from outside as provided in the master kit.
  • The extra burden of alcohol from wash buffer (1 needs 16 ml while wash buffer 2 needs 160 ml) to extraction step ( 560 µl for each test).
