Morphological Characteristics of Cryptococcus neoformans var. grubii in Lactophenol Cotton Blue Preparation

Cryptococcus neoformans var. grubii in a Lactophenol Cotton Blue (LPCB) preparation:

When examining a culture of Cryptococcus neoformans var. grubii  (Serotype A) Strainsin a Lactophenol Cotton Blue (LPCB) preparation, we can observe the following distinctive characteristics:

Appearance

Shape: The yeast cells are typically round or oval.
Size: Cells are usually 4-10 micrometers in diameter.
Capsule: A prominent clear halo surrounds each yeast cell, indicating the presence of a polysaccharide capsule. The capsule does not stain with LPCB, making it appear as a clear zone around the cell.
Color: The cell bodies stain blue due to the LPCB stain, while the capsule remains unstained, creating a clear halo effect.

Budding

Narrow-Based Budding: Cells exhibit narrow-based budding, a characteristic feature of Cryptococcus species.

Additional Observations

Cluster Formation: Yeast cells may be seen singly or in small clusters.
Consistency: The preparation typically shows a uniform distribution of cells without significant clumping.

Key Diagnostic Features

Polysaccharide Capsule: The presence of a large, clear capsule is a hallmark of Cryptococcus neoformans var. grubii ( the reduction of .capsule expression due to lacking host cell interaction being invitro condition as shown in the below footage)
Narrow-Based Budding: This type of budding is distinctive and helps differentiate it from other yeast species ( as in the figure below).
